"Micropropagation of peach rootstocks: research at Embrapa Clima Temperado, Brazil"

RESUMEN (ABSTRACT)
"Although there are numerous studies on micropropagation of peach rootstocks, in Brazil there is no definition of a protocol that is commercially applicable for selected genotypes. In the Rio Grande do Sul state, the largest peach producer in the country, accounting for 50.59% of production, Peach Tree Short Life - PTSL has been a major agronomic problem faced by growers. One research line in progress at Embrapa Clima Temperado aims to develop a protocol for micropropagation of several Prunus spp. access to obtain high healthy clonal rootstocks. This study aimed to establish in vitro peach rootstocks on different culture media. Nodal segments of approximately 1 cm containing one bud were excised from shoots collected from 2-yr old stock-plants grown in a greenhouse. The explants from 33 peach genotypes (hybrids, Prunus persica selections and another Prunus species) were surface sterilized with 70% alcohol for 2 minutes and with sodium hypochlorite 1% for 10 minutes and inoculated in 1/2MS, MS, WPM, AP, B5 culture medium. Subsequently, they were placed in a growth chamber at 24 ± 2 ¼C, 16h-photoperiod and light intensity of approximately 2000 lux. After 30 days, it was found that the establishment, fungus and bacterial contamination, as well as oxidation percentage, varied according to genotype and culture medium used. The in vitro establishment of explants was up to 38%, whereas the percentage of fungus and bacterial contamination, and oxidation was up to 41%."
